The Pattern of Gene Amplification Is Determined by the Chromosomal Location of Hairpin-Capped Breaks

چکیده
DNA palindromes often colocalize in cancer cells with chromosomal regions that are predisposed to gene amplification. The molecular mechanisms by which palindromes can cause gene amplification are largely unknown. Using yeast as a model system, we found that hairpin-capped double-strand breaks (DSBs) occurring at the location of human Alu-quasipalindromes lead to the formation of intrachromosomal amplicons with large inverted repeats (equivalent to homogeneously staining regions in mammalian chromosomes) or extrachromosomal palindromic molecules (equivalent to double minutes [DM] in mammalian cells). We demonstrate that the specific outcomes of gene amplification depend on the applied selection, the nature of the break, and the chromosomal location of the amplified gene relative to the site of the hairpin-capped DSB. The rules for the palindrome-dependent pathway of gene amplification defined in yeast may operate during the formation of amplicons in human tumors.

Parametric Study of the Empty and Foam-Filled End-Capped Conical Tubes under quasi Static and Dynamic impact Loads
This paper investigates the parametric study of the empty and foam-filled end-capped tubes under quasi static and dynamic loadings. The numerical crash analysis of the empty and foam-filled tubes was performed using the explicit finite element code ABAQUS- explicit. Satisfactory agreements were generally achieved between the numerical and experimental results. In order to determine the crash be...
متن کاملEvaluation of Resistance to Fluoroquinolones and Its Relationship whit parC Gene Mutation in Klebsiella pneumoniae Clinical Isolates
Background & Objective: Klebsiella pneumoniae has received attention due to a wide range of diseases and antibiotic resistance. The resistance to fluoroquinolones in gram-negative bacteria is often due to chromosomal mutations in the gyr and par genes. This research aimed at investigating the pattern of fluoroquinolone resistance and its relation with a mutation in the parC gene among clinical i...
متن کاملInduction of Resistance to Potato Virus Y (PVY) Using Hairpin Construct of Coat Protein
Potato virus Y (PVY) is one of the most damaging viruses of potato plants which infecting most cultivars and causing significant yield and economical losses. The application of the concept of pathogen derived resistance opened new horizons for the development of virus-resistant plants. This research was carried out to study RNA silencing to engineered potato plants that are resistant to potato ...
متن کاملP-230: Analysis of TEX15 Expression in Testis Tissues of Severe Oligozoospermic and Non-Obstructive Azoospermic Men Referred to Royan Institute
Background: TEX15 is a novel protein that is required for chromosomal synapsis and meiotic recombination. Human TEX15 is located on chromosome 8(8p12 region) and expressed in testis and ovary, as is its mouse ortholog. Loss of TEX15 function in mice causes early meiotic arrest in males but not in females. Specifically, TEX15 deficient spermatocytes exhibit a failure in chromosomal synapsis. In ...
متن کاملمهار بیان ژن GFP به وسیله تداخل RNA (RNAi) در دودمان سلولی کارسینومای جنینی P19
Introduction: RNA interference (RNAi) is a phenomenon of gene silencing that uses double-stranded RNA (dsRNA), specifically inhibits gene expression by degrading mRNA efficiently. The mediators of degradation are 21- to 23-nt small interfering RNAs (siRNA). The use of siRNAs as inhibitors of gene expression has been shown to be an effective way of studying gene function in mammalian cells. Ai...
